This experience must include four 4 years of experience with each of the followingbr br CRISPRCas9 plasmid design, transfection of embryonic stem cells;br Screening of transfected embryonic stem cell clones by southern blot and PCR genotyping; and,br Screening and genotyping of mice progeny.br One 1 year of experience in handling, maintenance and propagation of the human and rodent malaria life cycles and in generation of transgenic P. falciparum and P. yoelii;br br 3 One 1 year of experience with each of the followingbr Efforts in designing an antimalarial whole parasite based vaccine including generation of genetically attenuated P. falciparum and P.yoelii vaccine candidates using CRISPRCas9 based gene editing tools; and,br Screening of genetically attenuated P. falciparum vaccine candidates in the human liver chimeric humanized FRG huHep mouse model, including assessment of liver stage attenuation and livertoblood stage transition of these vaccine candidates.br br Experience and education may be gained concurrently.
