Experience, which may be gained concurrently, must includebr br 4 years of experience in mammalian cell culture including various cancer cell lines like MDAMB231, MIAPaCa2, Panc1, SKNMC, HEK293T, SHSY5Y, U251, U87; cryopreservation for banking cell lines;br 2 years of experience in cell biology techniques including clonogenic assays, viability and cytotoxicity assays using CellTiterFluor and CellTiterGlo, apoptosis assay using Annexin VPI staining for Flow cytometry analysis by BD Accuri C6, small molecule efficacy and toxicity screening, mycoplasma testing, subcellular fractionation, and transient transfections of plasmids, siRNA, and antisense oligonucleotides;br 2 years of experience conducting molecular and biochemical assays including nucleic acid purification from cells and tissues, RTPCR and qPCR, protein isolation, protein quantification using BCA assay, and protein expression analyses using ELISA and western blotting;br 1 year of experience in drug resistant stable cell line generation, lentiviral transduction, maintaining spheroid and organoid cultures derived from breast and prostate cancer patient tumors;br 1 year of experience in specimen preparation and analysis by immunohistochemistry;br 1 year of experience in characterizing mitochondrial bioenergetics by mitostress and electron flow assay using Agilent Seahorse XF96, amplex red assay to measure H2O2 activity to study ROS, studying ROS toxicity using NADPNADPH assay, GSHGSSG assay and protein carbonylation assay;br 1 year of experience in in vivo studies including mice anesthesia, injections, surgeries for implanting hormonal pellets, xenografting for tumor induction in breasts of female mice, bioluminescence image analysis for mice randomization for drug response studies, euthanize and dissect mice for extracting tumors, enzymatic dissociation of tumor to isolate and purify tumor cells for tumor microenvironment studies.
